Squid Giant Axon

The squid giant axon is an unusually large nerve fiber that transmits electrical signals and has become a foundational model for studying neuronal excitability. Its wide diameter permits direct insertion of electrodes, allowing researchers to measure membrane voltage as voltage-gated sodium and potassium channels open and close during an action potential, producing rapid depolarization followed by repolarization. Experiments with this axon established key principles of ion conductance, membrane potential, and action-potential propagation that underpin the Hodgkin-Huxley model. In neuroscience, the squid giant axon continues to support research in electrophysiology, neural signaling, and the cellular mechanisms underlying information transmission.

The method outlines the procedure by which the Hawaiian bobtail squid, Euprymna scolopes and its bacterial symbiont, Vibrio fischeri, are raised separately and then introduced to allow for specific colonization of the squid light organ by the bacteria. Colonization detection by bacterially-derived luminescence and by direct colony counting are described.

The Giant Fiber System is a simple neuronal circuit of adult Drosophila melanogaster containing the largest neurons in the fly. We describe the protocol for monitoring synaptic transmission through this pathway by recording post synaptic potentials in dorsal longitudinal (DLM) and tergotrochanteral (TTM) muscles following direct stimulation of the Giant Fiber interneurons.
