共聚焦荧光显微镜是一种专门用于定位细胞或组织样本中目标蛋白或抗原的成像技术,其原理是用荧光染料标记的抗体对抗原进行标记,并检测产生的荧光信号。该技术通过在物镜焦平面处设置两个针孔,实现了比宽场荧光显微镜更高的空间分辨率,因此得名“共聚焦”。该技术使用户能够在亚细胞水平上观察染色情况,例如区分细胞膜表面染色与细胞内染色。

共聚焦显微镜遵循与经典荧光显微镜类似的基本原理。光源(通常为共聚焦显微镜中的激光)发出的光束被二向色镜反射,并通过物镜聚焦到样品上。此光线激发荧光基团发射出不同波长的光,该发射光经物镜和二向色镜返回,最终到达相机或目镜。
