在大多数免疫学研究中,检测免疫细胞的增殖是一个关键步骤,常用基于CFSE荧光染料的方法进行检测。细胞的正常分裂对免疫细胞至关重要,因为它调控着免疫应答的强度和特异性。例如,T细胞通过增殖来识别并杀伤癌细胞,而B细胞则通过细胞分裂产生特异性抗体。CFSE检测的基本原理是用绿色荧光染料CFSE对细胞进行染色,该染料可进入活细胞并稳定地与胞内蛋白质结合,从而实现永久性标记。因此,当含有染料的亲代细胞分裂时,每个子代细胞将获得亲代细胞一半的荧光强度。
