Dit protocol beschrijft een eenvoudige benadering voor lijm-tape-gebaseerde steekproef van tomaten en andere verse producten oppervlakken, gevolgd door on-tape fluorescentie in situ hybridisatie (FISH) voor snelle cultuur-onafhankelijke detectie van Salmonella spp. Cell-tapes opgeladen kan ook het gezicht naar beneden worden geplaatst op selectieve agar voor vaste-fase verrijking voorafgaand aan de detectie. Als alternatief kan een laag-volume vloeistof verrijkingen (vloeistofoppervlak miniculture) worden uitgevoerd op het oppervlak van de tape in niet-selectieve bouillon, gevolgd door FISH en analyse via flowcytometrie. Om te beginnen, steriele zelfklevende tape is in contact gebracht met verse producten, wordt lichte druk toegepast, en de tape is verwijderd, fysiek extraheren microben aanwezig zijn op deze oppervlakken. Tapes zijn gemonteerd sticky-kant naar boven op glas microscoop dia's en de bemonsterde cellen gefixeerd met 10% formaline (30 min) en gedehydrateerd met behulp van een ethanolreeks (50, 80, en 95%, 3 minuten elke concentratie). Vervolgens cel geladen tapes zijn gespot met een buffer met een Salmonella-gerichte DNA-probe cocktail en gehybridiseerd gedurende 15 – 30 min bij 55 ° C, gevolgd door een korte spoeling in een wasbuffer om ongebonden probe te verwijderen. Aanhanger, zijn FISH-gelabelde cellen vervolgens tegengekleurd met de DNA-kleurstof 4 ',6-diamidino-2-phenylindole (DAPI) en de resultaten worden bekeken met behulp van fluorescentie microscopie. Voor vaste-fase verrijking, worden cel-betalen tapes face-down geplaatst op een geschikte selectieve agar oppervlak en geïncubeerd om in situ groei van Salmonella microkolonies, gevolgd door FISH en microscopie zoals hierboven beschreven. Voor vloeistofoppervlak miniculture, zijn cel geladen tapes geplaatst kleverige kant omhoog en een siliconen perfusie kamer wordt toegepast, zodat de tape en het objectglaasje vormen de bodem van een waterdichte kamer waarin een klein volume (≤ 500 pi) van trypticasesoja bouillon (TSB) is geïntroduceerd. De inlaat poorten zijn afgesloten en de kamers zijn geïncubeerd bij 35 – 37 ° C, waardoor de groei-gebaseerde amplificatie van de tape-geëxtraheerde microben. Na de incubatie, inlaatpoorten zijn onverhard, cellen zijn vrijstaand en gemengd met krachtig heen en weer pipetteren, geoogst via centrifugeren en gefixeerd in 10% neutraal gebufferde formaline. Tot slot, worden monsters gehybridiseerd en onderzocht via flowcytometrie om de aanwezigheid van Salmonella spp onthullen. Eenvoudige en snelle methodes voor de detectie van pathogenen op de productie van oppervlakken kan helpen verzachten door voedsel overgedragen ziekten door het verstrekken van tijdige en bruikbare data.
