Fluorescent Total Protein Stain

Fluorescent total protein stain is a biochemical method for visualizing and estimating the abundance of proteins in complex samples, commonly after separation by gel electrophoresis. Fluorescent dyes bind broadly to proteins through interactions with amino acid side chains, and the bound dye emits light at a characteristic wavelength when excited by an imaging system; signal intensity can reflect relative protein quantity within an appropriate linear range. This approach enables rapid assessment of sample composition, electrophoretic separation, loading consistency, and transfer efficiency, while providing greater sensitivity and compatibility with multiplex fluorescence workflows than many traditional colorimetric stains.

The stain-free technology allows researchers to visualize protein separation quality, to verify the transfer efficiency, and most importantly, to validate the change in the protein of interest using total protein quantification as a reliable loading control.

Here, we describe a detailed protocol outlining a new fluorescent staining technique for total protein detection in polyacrylamide gels. The protocol utilizes a silver ion-specific fluorescence turn-on probe, which detects Ag+-protein complexes, and eliminates certain limitations of traditional chromogenic silver stains.
