3d Wounded Cornea Model

A 3D wounded cornea model is a laboratory-engineered tissue system that reproduces the cornea’s layered structure and injury response, enabling controlled study of repair in a physiologically relevant setting. It typically combines corneal cells with a biomaterial or extracellular matrix scaffold, then introduces a defined wound to monitor cell migration, proliferation, matrix remodeling, and re-epithelialization under controlled culture conditions. In bioengineering, these models support evaluation of biomaterials, therapeutics, and regenerative strategies while reducing reliance on animal studies. They can also reveal how tissue architecture and biochemical signals influence healing, helping guide the development of corneal implants and personalized treatment approaches.

The cornea is unique in that it lacks vascular tissues. However, robust blood vessel growth and survival can be induced in the cornea by potent angiogenic factors. Therefore, the cornea can provide with us a valuable tool for angiogenic studies. This protocol demonstrates how to perform the mouse model of cornea pocket assay and how to assess the angiogenesis induced by angiogenic factors using this model.

An example of adenoviral gene therapy in the human diabetic organ-cultured corneas is presented towards the normalization of delayed wound healing and markedly reduced epithelial stem cell marker expression in these corneas. It also describes the optimization of this process in stem cell-enriched limbal epithelial cultures.
