Cell Barrier Function

Cell barrier function is the ability of a layer of cells to regulate the movement of substances between biological compartments while maintaining tissue integrity. In epithelial and endothelial tissues, cell-cell junctions, membrane transporters, and selective permeability work together to control passage based on properties such as size, charge, and solubility. Bioengineering approaches use cultured cells, biomaterials, and microfluidic platforms to model barriers found in organs including the intestine, kidney, lung, and blood-brain interface. These systems support studies of transport, disease, drug delivery, and toxicity, while helping researchers develop more physiologically relevant alternatives to conventional cell cultures and animal models.

This protocol reviews Electric Cell-substrate Impedance Sensing, a method to record and analyze the impedance spectrum of adherent cells for the quantification of cell attachment, proliferation, motility, and cellular responses to pharmacological and toxic stimuli. Detection of endothelial permeability and assessment of cell-cell and cell-substrate contacts are emphasized.

The aim of the protocol is to present an optimized procedure for the establishment of an in vitro blood-brain barrier (BBB) model based on primary porcine brain endothelial cells (pBECs). The model shows high reproducibility, high tightness, and is suitable for studies of transport and intracellular trafficking in drug discovery.

Functional Neuroimaging Using Ultrasonic Blood-brain Barrier Disruption and Manganese-enhanced MRI

A technique is described for broadly opening the blood-brain barrier in the mouse using microbubbles and ultrasound. Using this technique, manganese can be administered to the mouse brain. Because manganese is an MRI contrast agent that accumulates in depolarized neurons, this approach enables imaging of neuronal activity.
