Fibroblast Isolation

Fibroblast isolation is the process of obtaining fibroblasts from tissue or cell mixtures, providing a defined cell population for studying extracellular matrix production, tissue repair, and engineered constructs. The method typically combines tissue mincing with enzymatic digestion, using agents such as collagenase or dispase to disrupt the surrounding matrix, followed by filtration, centrifugation, and culture conditions that support fibroblast attachment and expansion. In bioengineering, isolated fibroblasts help generate tissue models, investigate cell–matrix interactions, and evaluate biomaterials for wound healing and regenerative applications. Consistent isolation and characterization improve reproducibility when comparing cell sources, culture conditions, and engineered tissue outcomes.

Fibroblast behavior underlies a spectrum of clinical entities, but they remain poorly characterized, largely due to their inherent heterogeneity. Traditional fibroblast research relies upon in vitro manipulation, masking in vivo fibroblast behavior. We describe a FACS-based protocol for the isolation of mouse skin fibroblasts that does not require cell culture.

Isolation of Rat Portal Fibroblasts by In situ Liver Perfusion

A technique for isolating portal fibroblasts from rat liver is described. Livers are perfused and digested in situ with collagenase, followed by ex vivo digestion of the liver slurry and size selection of cells. This method provides a pure population of portal fibroblasts without the need for passage in culture.

Here, a protocol to isolate and establish primary fibroblast/myofibroblast (MF) cultures from frozen gastric, small intestinal, and colonic tissue-yielding cells with a MF phenotype-is presented. These cells express CD90, α-SMA and vimentin. MFs can be used for a variety of functional assays including enzymatic activity and cytokine production.
