Human Aortic Endothelial Cells

Human aortic endothelial cells are specialized cells that line the inner surface of the aorta, where they form a selective interface between blood and the vessel wall. They regulate permeability, inflammation, coagulation, and vascular tone by sensing biochemical signals and hemodynamic forces such as fluid shear stress, which can stimulate signaling pathways and nitric oxide production. In bioengineering, these cells help evaluate vascular biomaterials, model endothelial dysfunction, and study how engineered vessels respond to blood flow. Their use supports the development of tissue-engineered blood vessels, cardiovascular disease models, and in vitro platforms for testing drugs and medical devices.

Isolation and Primary Culture of Mouse Aortic Endothelial Cells

The vascular endothelial cells play a significant role in many important cardiovascular disorders. This article describes a simple method to isolate and expand endothelial cells from the mouse aorta without using any special equipment. Our protocol provides an effective means of identifying mechanisms in endothelial cell physiopathology.

Restenosis following cardiovascular procedures (bypass surgery, angioplasty, or stenting) is a significant problem reducing the durability of these procedures. An ideal therapy would inhibit smooth muscle cell (VSMC) proliferation while promoting regeneration of the endothelium. We describe a model for simultaneous assessment of VSMC proliferation and endothelial function in vivo.

The goal of this protocol is to isolate endothelial progenitor cells from umbilical cord blood. Some of the applications include using these cells as a biomarker for identifying patients with cardiovascular risk, treating ischemic diseases, and creating tissue-engineered vascular and heart valve constructs.

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.
