Antibody Immobilization

Antibody immobilization is the attachment of antibodies to a solid surface or support while preserving their ability to recognize specific molecular targets. It works through covalent coupling or noncovalent adsorption, with the choice of chemistry and antibody orientation influencing antigen accessibility, binding strength, and stability. Immobilized antibodies form the basis of immunoassays, affinity purification, biosensors, and other biological techniques that isolate or detect proteins, cells, and small molecules. Controlling surface density, spacer design, and coupling conditions can improve assay sensitivity, reduce nonspecific binding, and support repeated use in research and diagnostic workflows.

Determining Binding Affinity (KD) of Radiolabeled Antibodies to Immobilized Antigens

Here, a method is described to determine the binding affinity (KD) of radiolabeled antibodies to immobilized antigens. KD is the equilibrium dissociation constant that can be determined from a saturation binding experiment by measuring the total, specific, and nonspecific binding of a radiolabeled antibody at various concentrations to its antigen.

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.
