Specific Dna Amplification

Specific DNA amplification is a molecular biology technique that selectively increases the amount of a chosen DNA sequence from a complex sample, enabling sensitive analysis of genetic material. It relies on sequence-specific primers that flank the target region; repeated cycles of denaturation, primer annealing, and extension by a thermostable DNA polymerase produce exponential enrichment of the intervening sequence. By limiting amplification to the intended target, the method supports accurate detection and characterization of DNA. It is widely used in biological techniques for pathogen identification, genotyping, mutation analysis, cloning, and preparation of samples for sequencing.

Amplification, Next-generation Sequencing, and Genomic DNA Mapping of Retroviral Integration Sites

We describe a protocol for amplifying retroviral integration sites from the genomic DNA of infected cells, sequencing the amplified virus-host junctions, and then mapping these sequences to a reference genome. We also describe techniques to quantify the distribution of integration sites relative to various genomic annotations using BEDTools.

Visualization of Mitochondrial DNA Replication in Individual Cells by EdU Signal Amplification

We developed a sensitive technique to label newly synthesized mitochondrial DNA (mtDNA) in individual cells in order to study mtDNA biogenesis. The technique combines the incorporation of EdU together with a tyramide signal amplification (TSA) protocol to visualize mtDNA replication within subcellular compartments of neurons.

Diagnosis of Eimeria infection in chickens remains demanding. Parasite morphology- and host pathology-led approaches are commonly inconclusive, while molecular approaches based on PCR have proven demanding in cost and expertise. The aim of this protocol is to establish loop-mediated isothermal amplification (LAMP) as a straightforward molecular diagnostic for eimerian infection.
