Buccal Swab Sampling

Buccal swab sampling is a noninvasive method for collecting cells from the inside of the cheek, providing biological material for genetic and cellular analysis. A sterile swab is gently rubbed against the buccal mucosa to dislodge epithelial cells, which are then preserved and processed to extract DNA or examine cellular characteristics. In biology, this technique supports genotyping, human identification, population studies, and research on inherited traits or disease-associated variants. Because it is painless, inexpensive, and easy to perform, buccal sampling offers a practical alternative to blood collection when only small amounts of genomic material are required.

In the present protocol, a urethral stricture induction was developed in Wistar rats, followed by urethral reconstruction with a buccal mucosa graft. A retrograde urethrogram and laser Doppler assessment were performed, validating urethral reconstruction (after stricture formation) and graft placement.
