Cerebrospinal Fluid Collection

Cerebrospinal fluid collection is the process of obtaining cerebrospinal fluid (CSF), the clear liquid that surrounds and protects the brain and spinal cord, for laboratory analysis and clinical assessment. Most commonly, clinicians perform a lumbar puncture by inserting a sterile needle into the subarachnoid space of the lower spine, where CSF pressure can be measured and fluid collected into designated tubes. Analysis of CSF composition, including cells, proteins, glucose, and microorganisms, supports evaluation of infections, bleeding, inflammation, and other nervous system disorders. In biology and biomedical research, CSF collection also helps investigate neurological disease mechanisms and assess biomarkers or treatment responses.

CSF production, circulation, and absorption are tightly regulated processes essential for the brain and spinal cord to function properly. This protocol describes an improved technique for the abundant collection of cerebrospinal fluid (CSF) with no contamination from blood. With greater sample collection and purity, more analyses can be performed using CSF to further our understanding of diseases that affect the brain and spinal cord.

A method for mass spectrometric analysis of endogenous peptides in human cerebrospinal fluid (CSF) is presented. By employing molecular weight cut-off filtration, chromatographic pre-fractionation, mass spectrometric analysis and a subsequent combination of peptide identification strategies, it was possible to expand the known CSF peptidome nearly ten-fold compared to previous studies.
