Detrusor Muscle

Detrusor muscle is the layer of smooth muscle forming most of the urinary bladder wall, where it controls bladder filling and emptying. During storage, sympathetic signaling and compliant smooth-muscle properties allow the bladder to expand at relatively low pressure; when voiding begins, parasympathetic nerves release acetylcholine, activating muscarinic receptors and coordinated detrusor contraction, while the urethral outlet relaxes. This system supports continence and efficient urination, and its activity is studied through bladder physiology and urodynamic testing. Abnormal detrusor overactivity, impaired contraction, or remodeling can contribute to urgency, incontinence, or urinary retention, making the muscle important in research on lower urinary tract disorders and their treatment.

The detrusor-free bladder model enables direct access to the suburothelium to study local mechanisms for regulation of biologically active mediator availability in suburothelium/lamina propria during storage and voiding of urine. The preparation closely resembles filling of an intact bladder and allows pressure-volume studies to be performed without systemic influences.

Preparation and Utilization of Freshly Isolated Human Detrusor Smooth Muscle Cells for Characterization of 9-Phenanthrol-Sensitive Cation Currents

We describe a method for preparation of the single freshly isolated detrusor smooth muscle cells from human urinary bladder specimens employing a two-step enzymatic procedure. The obtained viable DSM cells can be studied by various single cell techniques including the described amphotericin-B patch-clamp electrophysiology to reveal physiological and pharmacological properties.

We present a method for establishing a detrusor underactivity model by conus medullaris transection in rats. Detrusor underactivity was successfully stimulated in these animals. The model can be used for studying urinary tract function.
