Efferent Ducts

Efferent ducts are a network of small tubules that connect the rete testis to the epididymis, forming an essential passage in the male reproductive system. Their ciliated epithelial cells generate fluid currents that propel sperm, while nonciliated cells reabsorb much of the surrounding fluid and help concentrate the sperm before they enter the epididymis. Studying efferent ducts clarifies how sperm transport and maturation begin after production in the seminiferous tubules. Their structure and function are also important in reproductive biology, including research on infertility, hormonal regulation, and disorders that obstruct sperm movement.

Cannulation of the Mouse Submandibular Salivary Gland via the Wharton's Duct

A protocol for the cannulation of the mouse submandibular salivary gland via the Wharton's duct is described. For this experiment, the trypan blue solution is used as a dyer to demonstrate how this technique effectively delivers infusions into the targeted gland, and to suggest the reliability of this new approach as a potential clinical drug/cell therapy for the regeneration of salivary glands.

We present a method for isolation and culture of the mouse Wolffian duct (WD). We also demonstrate a detailed procedure for whole mount immunostaining of cultured/freshly isolated WDs with fluorescently tagged antibodies. Together, these techniques enable the study of WD development, coiling, and differentiation.
