Fluorescent Staining

Fluorescent staining is a biological labeling method that uses fluorescent dyes or tags to make specific cells, structures, or molecules visible under a fluorescence microscope. Fluorophores absorb light at one wavelength and emit light at a longer wavelength, while chemical stains or fluorescently labeled antibodies bind selected cellular components to produce targeted signals. In biology, this approach supports the localization and identification of proteins, organelles, nucleic acids, and other structures in fixed or living specimens. By revealing spatial distribution and changes in cellular organization, fluorescent staining aids microscopy, disease research, developmental studies, and analysis of cellular processes.

A method for fluorescent staining of fixed biological material with the specific DNA label methyl green is described. Methyl green is used in a diluted aqueous solution and is very resistant to photobleaching. Its far-red emission allows for deep specimen imaging, making it particularly adequate for whole embryos.

Here, we describe a detailed protocol outlining a new fluorescent staining technique for total protein detection in polyacrylamide gels. The protocol utilizes a silver ion-specific fluorescence turn-on probe, which detects Ag+-protein complexes, and eliminates certain limitations of traditional chromogenic silver stains.

Laparoscopic S7 Hepatectomy with Positive Fluorescence Staining
