Gel Loading Dye

Gel loading dye is a laboratory reagent added to DNA, RNA, or protein samples before gel electrophoresis to prepare them for loading and monitor their separation. It typically contains a dense substance, such as glycerol or sucrose, that helps the sample sink into the wells, along with colored tracking dyes that migrate through the gel at predictable rates. During electrophoresis, these dyes provide a visual indication of how far the run has progressed without necessarily staining the biomolecules themselves. Gel loading dye improves sample handling, reduces loss from wells, and supports accurate analysis of nucleic acid or protein band patterns in biology research and teaching.

Targeted-esterase induced dye loading (TED) supports the analysis of intracellular calcium store dynamics by fluorescence imaging. The method bases on targeting of a recombinant Carboxylesterase to the endoplasmic reticulum (ER), where it improves the local unmasking of synthetic low-affinity Ca2+ indicator dyes in the ER lumen.

Microelectrode-Mediated FM Dye Loading and Unloading in Drosophila Larvae Nerve Cells

In this video, Drosophila larvae nerve fibers are immersed in FM dye, which binds to cell membrane lipids. Suction microelectrode and electrical pulses stimulate the endocytosis process, incorporating the FM dye into newly formed endocytic vesicles. This method, which visualizes presynaptic stages of neurotransmission, is useful for studying synaptic function and neurotransmitter release.

Here, we describe a method for loading a calcium-sensitive dye through the frog nerve stump into the nerve endings. We also present a protocol for the recording and analysis of fast calcium transients in the peripheral nerve endings.
