Gene Deletion

Gene deletion is the removal or disruption of a defined DNA sequence so that a gene no longer produces its normal functional product, making it a powerful way to test gene function in biology. Researchers can create deletions through homologous recombination or targeted genome-editing systems such as CRISPR-Cas, which direct DNA cutting or replacement at selected genomic sites; cellular repair then joins the DNA ends or incorporates a designed template. Comparing mutant cells or organisms with unmodified controls can reveal effects on development, physiology, and disease-related pathways. Gene-deletion models also support functional genomics, pathway analysis, and evaluation of potential therapeutic targets.

Gene deletion mutants generated through homologous recombination are the gold standard for gene function studies. The OSCAR (One Step Construction of Agrobacterium-Recombination-ready-plasmids) method for rapid generation of deletion constructs is described. Agrobacterium mediated fungal transformation follows. Finally, a PCR based confirmation method of gene deletions in fungal transformants is presented.
