Hcv Replicon

An HCV replicon is an engineered hepatitis C virus RNA system that reproduces viral genome replication in cultured cells without producing infectious virus, making it a valuable model for studying HCV biology. Typically, the replicon retains the nonstructural proteins NS3 through NS5B, which assemble with host factors on intracellular membranes to copy the viral RNA, while structural genes are replaced by a selectable marker or reporter gene. Researchers use HCV replicons to measure RNA replication, analyze viral protein function, and screen antiviral compounds, including direct-acting drugs. These systems support mechanistic studies and safer, more controlled investigations of hepatitis C infection.

Two Methods of Heterokaryon Formation to Discover HCV Restriction Factors

We describe two methods for conditional trans-complementation of hepatitis C virus (HCV) assembly and the completion of the full viral life cycle, which rely on heterokaryon formation. These techniques are suitable to screen for cell lines that express dominant restriction factors, which preclude production of infectious HCV progeny.

The human lactoferrin (hLF) is a component of the immune system. In this study, immunofluorescence assays are used to demonstrate both the hepatocellular uptake of hLF and a qualitative reduction in the hepatitis C virus replication upon treatment with hLF.
