Imagej Subtraction Technique

The ImageJ subtraction technique is a digital image-processing method that removes background or reference-image intensity from microscopy data, helping reveal biologically meaningful signals. It operates by subtracting pixel values individually, often using a background image or a defined intensity value, so that shared features are reduced while differences remain; image bit depth and pixel-value limits influence the result. In biology, subtraction can improve contrast in fluorescence and bright-field images, correct uneven illumination, and isolate changes between experimental conditions. The processed images support more reliable visualization and quantitative measurements of cellular structures, molecular signals, and treatment effects.
