Immunosuppressive Drugs

Immunosuppressive drugs are medications that reduce or regulate immune-system activity, helping prevent harmful immune responses in transplantation, autoimmune disease, and inflammatory disorders. They work through different mechanisms, including limiting lymphocyte activation and proliferation, suppressing cytokine production, or reducing the activity of immune cells that recognize tissues as foreign or damaged. In biology and medicine, these drugs support organ and tissue graft survival and help control diseases caused by excessive immune activity, but their effects can also increase susceptibility to infection and other complications. Understanding their targets and cellular actions guides treatment selection, monitoring, and the development of more selective therapies.

Using optimized human peripheral blood mononuclear cell (PBMC) ex vivo assays, we showed that a combustible tobacco product preparation markedly suppresses receptor-mediated intracellularly secreted cytokines and cytolytic ability of effector PBMCs. These rapid assays may be useful in product evaluation and understanding the potential long-term effects of tobacco exposure.

T-lymphocyte mitogenesis is accompanied by blastogenic transformation, whereupon the cell volume enlarges before cell division. Here, we describe a method to quantify blastogenesis in T lymphocytes using an automated cell counter with the capability of measuring cell diameters.
