Lacz Lacy Laca

LacZ, LacY, and LacA are the three structural genes of the bacterial lac operon, a classic genetic system for regulating lactose utilization in Escherichia coli. When lactose is available and preferred glucose is scarce, the inducer allolactose reduces LacI repressor binding, allowing transcription of these genes. LacZ encodes β-galactosidase, which cleaves lactose and produces the inducer; LacY encodes a permease that transports lactose into the cell; and LacA encodes a transacetylase involved in processing related compounds. This coordinated regulation illustrates inducible gene expression, metabolic control, and the relationship between environmental conditions and bacterial phenotype.

Prokaryotes can control gene expression through operons—DNA sequences consisting of regulatory elements and clustered, functionally related protein-coding genes. Flow cytometry is a powerful tool allowing for the isolation and study of specific cell populations. This protocol describes steps for isolating LacZ-expressing cells from cochlear tissues from neonatal transgenic mice. Dissociated cochlear cells were labeled using fluorescent-conjugated substrates of β-galactosidase prior to separation via flow cytometry.
