Lipid Envelope Disruption

Lipid envelope disruption is the breakdown of the lipid bilayer that surrounds certain viruses, cells, or membrane-bound particles, compromising their structural integrity and function. It occurs when detergents, solvents, changes in temperature or pH, or physical forces disturb interactions among phospholipids and membrane proteins, causing leakage, fragmentation, or loss of envelope organization. In biology, this process helps explain how enveloped viruses are inactivated and how membrane-associated components can be released for analysis. Controlled disruption is also used in sample preparation, antiviral research, vaccine development, and studies of membrane structure, stability, and transport.

This article describes a method to visualize formation of an HIV-1 envelope-induced virological synapse on glass supported planar bilayers by total internal reflection fluorescence (TIRF) microscopy. The method can also be combined with immunofluorescence staining to detect activation and redistribution of signaling molecules that occur during HIV-1 envelope-induced virological synapse formation.

Outer Layers of the Cell Envelope

2025

The outermost layers of prokaryotic cells play a critical role in their survival, virulence, and interaction with the environment. These layers, often composed of polysaccharides, polypeptides, or proteins, form protective and adhesive structures that vary in organization and function.Capsules and Slime LayersCapsules are highly organized, tightly bound layers that firmly attach to the bacterial cell wall. Capsules are usually made of polysaccharides, though some are made of polypeptides. These...

Conformational Evaluation of HIV-1 Trimeric Envelope Glycoproteins Using a Cell-based ELISA Assay

Understanding viral surface antigens conformations is required to evaluate antibody neutralization and guide the design of effective vaccine immunogens. Here we describe a cell-based ELISA assay that allows the study of the recognition of trimeric HIV-1 Env expressed at the surface of transfected cells by specific anti-Env antibodies.

Determination of Total Lipid and Lipid Classes in Marine Samples

This protocol is for the determination of lipids in seawater and biological specimens. Lipids in filtrates are extracted with chloroform or mixtures of chloroform and methanol in the case of solids. Lipid classes are measured by rod thin-layer chromatography with flame ionization detection and their sum gives the total lipid content.
