Live Cell Imaging

Live cell imaging is the visualization of living cells over time, allowing researchers to observe dynamic biology rather than relying only on fixed-cell snapshots. Using techniques such as phase-contrast or fluorescence microscopy, researchers capture time-lapse images while maintaining cells under suitable environmental conditions; fluorescent labels can reveal specific proteins, organelles, or cellular events. In biology, these measurements help characterize cell division, migration, morphology, intracellular transport, and responses to stimuli or treatment. Because the same cells can be followed longitudinally, live cell imaging links cellular behaviors to their timing and sequence, supporting quantitative analysis of development, disease mechanisms, and therapeutic effects.

Live Cell Imaging during Mechanical Stretch

A novel imaging protocol was developed using a custom motor-driven mechanical actuator to allow the measurement of real time responses to mechanical strain in live cells. Relevant to mechanobiology, the system can apply strains up to 20% while allowing near real-time imaging with confocal or atomic force microscopy.

Cell Death Associated with Abnormal Mitosis Observed by Confocal Imaging in Live Cancer Cells

The cytotoxic activity of the phenanthridine PJ-34 in cancer cells undergoing mitosis was documented in real time by live confocal imaging. PJ-34 eradicated human breast cancer MDA-MB-231 cells harboring extra-centrosomes in mitosis. Unlike normal bi-focal mitosis, the extra-centrosomes were not clustered in the two spindle poles in the presence of PJ-34.

4D Imaging of Protein Aggregation in Live Cells

Cellular viability depends on timely and efficient management of protein misfolding. Here we describe a method for visualizing the different potential fates of a misfolded protein: refolding, degradation, or sequestration in inclusions. We demonstrate the use of a folding sensor, Ubc9ts, for monitoring proteostasis and aggregation quality control in live cells using 4D microscopy.
