Local Calcium Signals

Local calcium signals are brief, spatially restricted increases in cytosolic Ca2+ that allow cells to regulate nearby molecular processes without raising calcium throughout the cell. They arise when calcium enters through plasma-membrane channels or is released from intracellular stores, then diffuses over short distances while buffers, pumps, and exchangers shape their amplitude and duration. These microdomains can activate calcium-sensitive proteins at specific sites, including those controlling neurotransmitter release, muscle contraction, membrane trafficking, and cell motility. Studying local calcium signals helps explain how cells convert electrical or chemical stimuli into precise responses and clarifies how disrupted calcium handling contributes to biological dysfunction.

This protocol outlines a simple method for analyzing calcium signals in plants generated by feeding hemipteran insects, such as aphids.
