Long-term Depression

Long-term depression (LTD) is a persistent reduction in synaptic strength, a form of biological plasticity that enables neural circuits to adjust their responses over time. In many excitatory neurons, prolonged low-frequency stimulation produces a modest calcium influx through NMDA receptors, activating signaling pathways that remove AMPA receptors from the postsynaptic membrane and reduce transmission efficiency. Studied in brain regions including the hippocampus and cerebellum, LTD helps refine developing circuits, support learning and memory, and balance long-term potentiation. Its mechanisms also provide insight into how disrupted synaptic plasticity may contribute to neurological and neurodevelopmental disorders.

In this video, a mouse cerebellar brain slice was used to investigate synaptic interactions between parallel fibers, climbing fibers, and Purkinje cells, which are essential for motor control. Stimulation of climbing fibers induced long-term depression (LTD) in Purkinje cells, weakening synaptic strength and resulting in reduced response to parallel fiber stimulation.

In some gene-manipulated animals, using a single protocol may fail to induce LTD in cerebellar Purkinje cells, and there may be a discrepancy between LTD and motor learning. Multiple protocols are necessary to assess LTD-induction in gene-manipulated animals. Standard protocols are shown.
