Matrigel Invasion Chamber

A Matrigel invasion chamber is a laboratory device used to measure the ability of cells to invade through a three-dimensional extracellular matrix, providing a model of tissue invasion. It typically contains a porous membrane coated with Matrigel, a basement membrane-like material, while cells are placed in an upper compartment and migrate toward a chemoattractant in the lower compartment. Cells with invasive properties degrade or penetrate the matrix, pass through the membrane pores, and can then be stained and counted. This assay supports studies of cancer metastasis, cell motility, extracellular matrix interactions, and the effects of potential anti-invasive treatments.

This video demonstrates how to maintain the growth of human embryonic stem cells (hESCs) in feeder cell-free conditions and how to continuously passage hESCs in feeder cell-free conditions. Confirmation of hESC pluripotency grown in feeder cell-free conditions by immunofluorescence microscopy is also demonstrated. Part 3 of 3.
