Membrane Blocking

Membrane blocking is a preparatory step in immunoblotting and other membrane-based assays that reduces nonspecific binding before target detection. After proteins are immobilized on a membrane, a blocking solution containing proteins such as bovine serum albumin or nonfat milk coats unoccupied binding sites, limiting unintended adsorption of antibodies and lowering background signal. Effective blocking improves the contrast between specific and nonspecific staining, helping researchers detect and quantify target proteins more reliably. The choice of blocking reagent and conditions can affect assay sensitivity, especially when antibodies or detection systems interact with components of the blocking solution.

Herein is described a robotic approach to high-throughput crystallization of membrane proteins in lipidic mesophases for use in structure determination using macromolecular X-ray crystallography. Three robots capable of handling the viscous and sticky protein-laden mesophase integral to the method are introduced.
