Autoimmune Disease

Autoimmune disease is a group of disorders in which the immune system mistakenly targets the body’s own cells, tissues, or organs, causing inflammation and potentially lasting damage. When immune tolerance breaks down, autoreactive T cells and antibodies recognize self-antigens as threats and activate inflammatory pathways that disrupt normal tissue function. These diseases can affect specific organs, as in type 1 diabetes, or multiple systems, as in systemic lupus erythematosus, producing diverse symptoms and complications. Studying autoimmune disease helps biologists understand immune regulation and supports the development of diagnostic biomarkers, immunomodulatory treatments, and strategies for preventing tissue damage.

Parasite Induced Genetically Driven Autoimmune Chagas Heart Disease in the Chicken Model

The inoculation of Trypanosoma cruzi in fertile eggs prior to incubation renders the parasite kDNA minicircle integration in embryo cells genome. Crossbreeding reveals the vertical transfer of the mutations to progeny. The kDNA integrates into coding regions at several chromosomes and the chickens die with an inflammatory autoimmune heart disease.

This manuscript describes the methods for induction and scoring of the experimental autoimmune encephalomyelitis (EAE) model, together with the assessment of immune cell distribution and mRNA cytokine levels in lymph nodes, spleen, blood and spinal cord using flow cytometry and quantitative PCR, respectively, at various disease phases.
