Nk Cell Enrichment

Natural killer (NK) cell enrichment is a cell-separation method that increases the proportion of NK cells in a mixed biological sample, enabling focused analysis of their roles in innate immunity. The process typically uses antibodies that recognize NK-cell surface markers to selectively retain desired cells or remove unwanted populations, followed by physical separation and recovery of an enriched fraction. Enriched NK cells support studies of cytotoxicity, cytokine production, immune regulation, infection, cancer biology, and cellular immunotherapy. By improving cell purity and consistency, the method helps researchers characterize NK-cell function and evaluate their therapeutic potential.

The goal of this study was to formulate technologies that allow for successful gene transduction in primary natural killer (NK) cells. The dextran-mediated lentiviral transduction of human or mouse primary NK cells results in higher gene expression efficiencies. This method of gene transduction will vastly improve NK cell genetic manipulation.
