Nude Mouse Graft

Nude mouse grafting is the transplantation of human or animal cells, tissues, or tumors into nude mice, immunodeficient animals widely used to study growth and treatment responses in vivo. These mice carry a hairless phenotype and an athymic condition that limits T-cell development, reducing cellular immune rejection and allowing xenografted material to establish and interact with host tissues. Researchers use nude mouse graft models to investigate tumor biology, angiogenesis, metastasis, and drug efficacy, while also evaluating tissue behavior in a living system. The approach links controlled laboratory manipulation with clinically relevant biological outcomes, although incomplete immunity and species differences must inform interpretation.

We describe protocols for our mouse graft arteriosclerois (GA) models which involve interposition of a mouse vessel segment into a recipient of the same inbred strain. By backcrossing additional genetic changes into the vessel donor, the model can assess the effect of specific genes on GA.

To improve our knowledge of cellular and molecular neotissue formation, a murine model of the TEVG was recently developed. The grafts were implanted as infrarenal vena cava interposition grafts in C57BL/6 mice. This model achieves similar results to those achieved in our clinical investigation, but over a far shortened time-course.
