Phospholipid Transport

Phospholipid transport is the movement of membrane lipids within and between cellular membranes, a process essential for building, maintaining, and remodeling membrane structure. Cells use vesicular trafficking, lipid transfer proteins, and specialized enzymes called flippases, floppases, and scramblases to distribute phospholipids between organelles and across bilayer leaflets, preserving or changing membrane asymmetry. This transport supports membrane biogenesis, organelle identity, cell signaling, and responses to stress. Studying phospholipid transport helps explain how cells regulate membrane composition and provides insight into disorders associated with defects in lipid metabolism, trafficking, and signaling.

Quantitative Flow Cytometry to Study Labeled Protein-Phospholipid Vesicle Interactions

In this video, we demonstrate the interaction between proteins and artificial phospholipid vesicles using quantitative flow cytometry. The binding of fluorescently-labeled proteins to fluorescently-labeled phospholipid vesicles increases the mean fluorescence intensity, and this increase is detected by a flow cytometer.

Extraction and Analysis of Microbial Phospholipid Fatty Acids in Soils

Phospholipid fatty acids provide information about the structure of soil microbial communities. We present methods for extraction from soil samples with a single-phase chloroform mixture, fractionation of extracted lipids using solid phase extraction columns, and methanolysis to produce fatty acid methyl esters, which are analyzed by capillary gas chromatography.
