Rodent Estrous Cycle

The rodent estrous cycle is the recurring sequence of reproductive changes that prepares female rodents for ovulation, mating, and potential pregnancy. Coordinated signaling between the hypothalamus, pituitary gland, and ovaries regulates ovarian hormone production, while changing estrogen and progesterone levels drive the characteristic stages of proestrus, estrus, metestrus, and diestrus. Researchers identify these stages through vaginal cytology and by monitoring reproductive behavior or hormone patterns. Understanding cycle timing is essential for interpreting fertility, pregnancy, reproductive endocrinology, and sex-related variation in laboratory studies, including research on development, physiology, disease, and drug effects.

Here, we describe how to identify the stage of the murine reproductive (proestrus, estrus, metestrus, or diestrus) by simple, non-invasive collection and cytological assessment of vaginal smear samples. We further describe how vaginal cytology reflects circulating hormonal levels underlying transition through the murine reproductive cycle.
