Sc1 Small Molecule

SC1 is a synthetic small molecule used in stem cell biology to support the self-renewal of mouse embryonic stem cells. It acts through two complementary pathways by inhibiting RasGAP, which modulates Ras signaling, and ERK1, a kinase in the MAPK pathway; this combination helps preserve an undifferentiated state, including under serum-free culture conditions without leukemia inhibitory factor. Researchers use SC1 to study pluripotency, signaling control, and the balance between self-renewal and differentiation. Its defined mechanism also makes it useful for developing culture systems and investigating how chemical regulation can replace or supplement growth-factor-based stem cell maintenance.

SC1 functions through dual inhibition of Ras- GAP and ERK1. We tested the function of SC1 in supporting mouse ES cell self-renewal in the absence of LIF and showed that SC1 is able to maintain self-renewal of mouse ES cell cultures.
