Slot Blot Technique

The slot blot technique is a membrane-based analytical method that detects and compares specific nucleic acids or proteins without separating them by gel electrophoresis. Samples are drawn through narrow slots onto a membrane, where target molecules become immobilized; labeled nucleic acid probes or antibodies then bind complementary sequences or epitopes, and washing removes nonspecific signal before detection. In biology, slot blots support rapid screening, semi-quantitative comparisons, and confirmation of target presence across many samples. The method is useful for analyzing gene expression, DNA or RNA abundance, and protein levels when throughput and direct sample comparison are more important than resolving molecules by size.

A simple slot blot method was developed for the quantification of influenza viral hemagglutinin and neuraminidase using universal antibodies targeting their most conserved sequences identified through bioinformatics analyses. This innovative approach may provide a useful alternative to quantitative determination of all viral hemagglutinin and neuraminidase.
