Soft Agar Overlay

Soft agar overlay is a microbiological technique that supports the growth, interaction, or detection of microorganisms within a semisolid layer of agar. A molten, low-percentage agar medium is mixed with cells, bacteria, or bacteriophages and poured over a solid agar base; as it cools, the soft matrix permits nutrient diffusion while restricting movement. This setup is widely used in bacteriophage plaque assays, where viral infection and bacterial lysis produce visible clear zones, and in assays of microbial growth or interactions. By converting microscopic biological events into measurable colonies or plaques, soft agar overlay provides a practical method for studying infection, replication, and microbial behavior.

Plaque assays are the gold standard for viral quantification, utilizing entrapping overlays on host cellular monolayers to determine viral titers. While various semisolid overlays have traditionally been used, here we demonstrate plaque techniques comparing semisolid overlays to a novel liquid microcrystalline cellulose among several families of viruses.
