Spindle Pole Body

The spindle pole body is a microtubule-organizing center found in fungi, where it serves as the functional counterpart of the centrosome and directs chromosome segregation. Embedded in the nuclear envelope, it duplicates before nuclear division and nucleates microtubules that form the mitotic or meiotic spindle, linking spindle architecture to accurate movement of chromosomes. Studying spindle pole bodies reveals how cells assemble bipolar spindles, coordinate nuclear-envelope organization, and control cell-cycle progression. Research in yeast and other fungi uses this structure to investigate conserved mechanisms of mitosis, meiosis, microtubule organization, and chromosome inheritance.

Here, we present a protocol for specific siRNA-mediated mRNA depletion followed by immunofluorescence analysis to evaluate meiotic spindle assembly and organization in mouse oocytes. This protocol is suitable for in vitro depletion of transcripts and functional assessment of different spindle and/or MTOC-associated factors in oocytes.
