Tissue-derived Tubuloids

Tissue-derived tubuloids are three-dimensional laboratory models formed from cells isolated from tubular tissues, providing a structured system for studying epithelial biology in a controlled setting. When tissue-derived cells are embedded in a supportive extracellular matrix and exposed to defined growth conditions, they proliferate and self-organize into tubule-like structures that can retain key features of the source tissue. These models help researchers investigate tissue development, transport functions, injury, and repair while reducing reliance on animal experiments. In biology and biomedical research, tissue-derived tubuloids also support disease modeling, drug response testing, and studies of personalized tissue function.

Generation of Human Kidney Tubuloids from Tissue and Urine

Human kidney tubuloid cultures represent a valuable in vitro model to study kidney physiology and disease. Tubuloids can be established from kidney tissue (healthy and diseased) as well as urine, the latter representing an easily obtainable and less invasive source of research material.

We present a protocol to isolate adipose tissue-derived microvascular fragments that represent promising vascularization units. They can be rapidly isolated, do not require in vitro processing and, thus, may be used for one-step prevascularization in different fields of tissue engineering.

This article outlines a versatile method to create cell-derived tissue rings by cellular self-assembly. Smooth muscle cells seeded into ring-shaped agarose wells aggregate and contract to form robust three-dimensional (3D) tissues within 7 days. Millimeter-scale tissue rings are conducive to mechanical testing and serve as building blocks for tissue assembly.

Quantification of Inflammatory Mediators in Infected Human Tissue-Derived Cells

This video demonstrates the quantification of inflammatory mediators during infection by incubating human lung-tissue-derived immune cells with Haemophilus influenzae. Flow cytometry is used to measure cytokine fluorescence in labeled lymphocytes, correlating with the production of pro-inflammatory cytokines in response to bacterial infection.

This protocol describes the isolation of pig adipose-derived stem cells (pADSC) from subcutaneous adipose tissues with examination of multipotency. The multipotent pADSC are used to delineate processes of adipocyte differentiation and study transdifferentiation into multiple cell lineages of mesodermal mesenchyme or further lineages of ectoderm and endoderm for regenerative studies.
