3d In Vitro Assay

3D in vitro assays are laboratory tests that study cells or tissues within a three-dimensional model, providing a more physiologically relevant context than flat, two-dimensional cultures. By allowing cells to grow in aggregates, scaffolds, or matrix-based environments, these assays reproduce spatial organization, cell-cell interactions, and gradients of nutrients, oxygen, and signaling molecules that influence behavior. In cancer research, tumor spheroids and organoid-based assays help evaluate proliferation, invasion, treatment response, and resistance while supporting studies of tumor biology and therapy development. Their controlled design can bridge basic cell experiments and more complex in vivo models, although model composition and culture conditions strongly affect interpretation.

We present a protocol to obtain cell-derived matrices rich in extracellular matrix proteins, using macromolecular crowders (MMC).
