Bispecific T Cell Engager

Bispecific T-cell engagers are engineered antibody-based molecules that connect T cells with cancer cells, offering a targeted way to direct immune-mediated tumor killing. Typically, one binding domain recognizes a tumor-associated antigen while the other binds CD3 on the T-cell receptor complex; this proximity forms an immune synapse that activates T cells to release cytotoxic molecules and destroy the engaged target cell, without requiring peptide presentation by major histocompatibility complex. In cancer research, these agents support studies of immunotherapy, antigen selection, T-cell activation, treatment resistance, and strategies to improve efficacy and safety across blood cancers and solid tumors.

An In Vivo Technique to Track Bispecific Antibody-Induced T Cell Trafficking to Tumors

This video demonstrates a method for tracking bispecific antibody-induced T cell trafficking to tumors in a mouse model. Luciferase-transduced effector T cells are injected into a mouse with a subcutaneous tumor cell xenograft. Bispecific antibodies are then introduced, binding to T cells and the tumor cells. The localization of the T cells is tracked using bioluminescent imaging, with the magnitude of the signal indicating the extent of T cell infiltration into the tumor.
