Hematopoietic Lineage

Hematopoietic lineage refers to the developmental pathways through which hematopoietic stem cells produce diverse blood and immune cell types, making it central to understanding blood formation and disease. In the bone marrow, self-renewing stem cells generate progressively restricted progenitors that branch into myeloid and lymphoid lineages, whose differentiation is guided by transcriptional programs and growth signals. Mapping these lineages helps researchers identify how normal blood development is disrupted by mutations, altered signaling, or abnormal self-renewal. In cancer research, lineage analysis supports leukemia classification, tracks tumor evolution, and informs studies of targeted therapies, treatment resistance, and the cellular origins of hematologic malignancies.

Phenotypic Analysis and Isolation of Murine Hematopoietic Stem Cells and Lineage-committed Progenitors

A method to analyse the distribution of bone marrow hematopoietic progenitors in flow cytometry as well as to efficiently isolate highly purified hematopoietic stem cells (HSCs) is described. The isolation procedure is essentially based on magnetic enrichment of c-Kit+ cells and cell sorting to purify HSCs for cellular and molecular studies.

The colony forming cell (CFC) assay is an in vitro assay in which hematopoietic progenitors form colonies in a semi-solid medium. A combination of colony morphology, cell morphology, and flow cytometry are used to assess the ability of the progenitors to proliferate and differentiate along the different hematopoietic lineages.
