Internal Carotid Injection

Internal carotid injection is a targeted technique that delivers tumor cells, drugs, tracers, or other experimental materials into the internal carotid artery, which supplies blood to the brain. After administration, the injected material follows arterial blood flow through the cerebral circulation, allowing researchers to examine how circulating cancer cells arrest in brain vessels or how treatments reach intracranial tumors. In preclinical cancer research, this approach supports models of brain metastasis, evaluation of tumor-cell interactions with the blood-brain barrier, and assessment of regionally directed therapies. Because delivery occurs close to the target tissue, the method can provide controlled experimental exposure while limiting dependence on broader systemic distribution.

Source: Lim, M., et al. Modeling Brain Metastasis by Internal Carotid Artery Injection of Cancer Cells. J. Vis. Exp. (2022)In this video, an anesthetized mouse undergoes a surgical procedure in which cancer cells are injected into the common carotid artery (CCA) to facilitate targeted delivery to the brain. The procedure involves precise suturing and manipulation of the carotid arteries to direct the injected cells toward the internal carotid artery (ICA), promoting tumor formation in the brain...

Detection of Internal Carotid Artery Narrowing Using Digital Subtraction Angiography

Position a human patient with internal carotid stenosis, a condition characterized by narrowing of the internal carotid artery, on the imaging table for digital subtraction angiography (DSA). Insert an angiocatheter into the common carotid artery at the fourth cervical vertebra level. Capture a pre-contrast X-ray image to record background structures like bones and soft tissues using X-ray irradiation. Administer an iodine-based contrast medium into the carotid artery. The contrast medium...

(2017)This video demonstrates a microsurgical procedure designed to facilitate the targeted infusion of a test substance into the brain via the internal carotid artery (ICA). The procedure involves precise suturing and vascular manipulation to temporarily restrict blood flow, ensuring controlled delivery. A capillary tube is inserted into...

Brain metastasis is a cause of severe morbidity and mortality in cancer patients. Most brain metastasis mouse models are complicated by systemic metastases confounding analysis of mortality and therapeutic intervention outcomes. Presented here is a protocol for internal carotid injection of cancer cells that produces consistent intracranial tumors with minimal systemic tumors.
