Ksfm Culture

KSFM culture is a laboratory method for growing keratinocytes and related epithelial cells in keratinocyte serum-free medium, providing controlled conditions for studying cell behavior. The formulation supplies essential nutrients and selected growth-promoting components without serum, reducing undefined biological factors that can influence proliferation, differentiation, and signaling. In cancer research, KSFM culture supports the maintenance and comparison of normal and transformed epithelial cells, helping investigators examine oncogenic changes, growth regulation, drug responses, and interactions with the cellular environment. Its defined conditions improve experimental reproducibility and can complement three-dimensional models and other approaches to epithelial tumor biology.

In this report we describe a method for the isolation and culture of the progenitor cell niche from the embryonic mouse kidney that can be used to study signaling pathways regulating stem/progenitor cells of the developing kidney. These cultured cells are highly accessible to small molecule and recombinant protein treatment, and importantly also to viral transduction, which allows efficient manipulation of candidate pathways.

This video demonstrates New culture, a method by which chick embryos are cultured outside the egg for up to 24 hr. This method enables one to study early development (primitive streak to 14 som.), a period corresponding to E7-9 in mouse. Applications of this technique include electroporation, in situ hybridization and immunohistochemistry.

Culture and Co-Culture of Mouse Ovaries and Ovarian Follicles

This protocol describes the primary culture/co-culture of mouse ovarian tissue, using ovaries from neonatal mice and individual ovarian follicles from prepubertal mice. The culture techniques support development in a highly physiological manner, allowing investigation of the effect of extrinsic agents on the ovary, and of interactions between ovarian follicles.
