Nude Mouse Model

A nude mouse model is a laboratory mouse with a congenital thymus defect that causes impaired T-cell development, making it useful for studying cancer under controlled biological conditions. In these athymic mice, human or mouse tumor cells can be implanted as xenografts because reduced adaptive immune rejection supports tumor establishment and growth, while other immune and physiological systems remain active. Researchers use nude mouse models to investigate tumor growth, invasion, metastasis, and responses to anticancer drugs or radiation. Their reproducibility supports preclinical evaluation, although findings may not fully predict outcomes in patients with intact immune systems.

In the pathogenesis of bone metastasis, angiogenesis is a crucial process and therefore represents a target for imaging and therapy. Here, we present a rat model of site-specific breast cancer bone metastasis and describe strategies to non-invasively image angiogenesis in vivo using magnetic resonance imaging, volumetric computed tomography and ultrasound.

Mycobacterium leprae, the causative agent of leprosy, does not grow in vitro. We describe an easy to follow protocol to prepare a bacillary suspension to ensure the maintenance of large quantities of M. leprae for a variety of applications. Protocols for propagation by mouse footpad inoculation, evaluation of viability, freezing and thawing bacillary stock are described in detail.
