Tumor Tissue Embedding

Tumor tissue embedding is a sample-preparation method that supports the preservation, orientation, and sectioning of tumor specimens for microscopic and molecular analysis. In the most common approach, tissue is fixed, dehydrated, cleared, and infiltrated with paraffin before cooling it into a firm block; frozen embedding uses a cryogenic medium to preserve samples without paraffin processing. The resulting blocks can be cut into thin sections for histopathology, immunohistochemistry, and assessment of tumor architecture, cellular features, and biomarker expression. Consistent embedding improves tissue quality and reproducibility, helping cancer researchers characterize disease, compare treatment responses, and investigate mechanisms of tumor development.

Methods to create human 3D tumor tissues as test systems are described. These technologies are based on a decellularized Biological Vascularized Scaffold (BioVaSc), primary human cells and a tumor cell line, which can be cultured under static as well as under dynamic conditions in a flow bioreactor.

Engineering Three-dimensional Epithelial Tissues Embedded within Extracellular Matrix

This manuscript describes a soft lithography-based technique to engineer uniform arrays of three-dimensional (3D) epithelial tissues of defined geometry surrounded by extracellular matrix. This method is amenable to a wide variety of cell types and experimental contexts and allows for high-throughput screening of identical replicates.
