Counter-electrode Preparation

Counter-electrode preparation is the process of selecting and treating the electrode that completes an electrochemical circuit and carries current during a measurement or reaction. In a three-electrode cell, the counter-electrode supports the complementary oxidation or reduction reaction to the process at the working electrode, so its conductivity, chemical stability, surface condition, and exposed area influence cell performance. Preparation may include choosing a suitable substrate, cleaning its surface, and applying a catalytic coating when required. Carefully prepared counter-electrodes improve current distribution, minimize contamination, and support reliable results in electroanalysis, electrosynthesis, corrosion studies, and energy-storage research.

We have developed an in vitro unfolded hippocampus which preserves CA1-CA3 array of neurons. Combined with the penetrating micro-electrode array, neural activity can be monitored in both the longitudinal and transverse orientations. This method provides advantages over hippocampal slice preparations as the propagation in the entire hippocampus can be recorded simultaneously.

Existing approaches for constructing chronically implantable peripheral nerve cuff electrodes for use in small rodents often require specialized equipment and/or highly trained personnel. In this protocol we demonstrate a simple, low-cost approach for fabricating chronically implantable cuff electrodes, and demonstrate their effectiveness for vagus nerve stimulation (VNS) in rats.
