Cd34 Sca-1 Expression

CD34 Sca-1 expression refers to the presence of two cell-surface markers used to identify and characterize stem and progenitor cell populations, particularly during hematopoietic development. CD34 is associated with immature blood-forming cells, while Sca-1, commonly analyzed in mouse models, helps distinguish stem and progenitor compartments; researchers typically assess both markers by fluorescent antibody labeling and flow cytometry. Their expression patterns can reveal changes in cellular identity, self-renewal potential, and differentiation during embryonic and postnatal development. In developmental biology, this marker combination supports the isolation of candidate hematopoietic stem cells and the study of how tissues establish and maintain blood-forming capacity.

This procedure describes a quick and easy workflow to introduce siRNA into difficult to transfect cell lines and follow gene expression by real-time PCR. Use of an automated cell counter, multi-well electroporation plate, and automated electrophoresis station provide quick and reliable results without the need for expensive robotic handling.

We have developed a lentiviral vector that possesses, in addition to the Tat-responsive LTR, the Rev-response element (RRE) that can regulate reporter gene expression in an HIV-1 Tat- and Rev-dependent fashion. The vector permits the specific detection of replicating HIV in living cells via the expression of GFP.
