Mouse Ovarian Tissue

Mouse ovarian tissue is the reproductive tissue that contains oocytes, follicles, and supporting somatic cells, making it an important model for studying mammalian reproductive development. Its function depends on coordinated folliculogenesis, in which oocytes develop within follicles while granulosa and theca cells provide structural support, nutrients, and hormone-responsive signaling. In developmental biology, mouse ovarian tissue helps researchers examine germ-cell specification, follicle formation, ovarian maturation, and interactions between germ cells and their surrounding microenvironment. Tissue culture and genetic analysis can further reveal how developmental signals influence fertility, reproductive aging, and ovarian disorders, supporting broader insights into mammalian organ development.

Transplantation Into the Mouse Ovarian Fat Pad

We describe an ovarian fad pad transplantation assay suitable for studies of normal and transformed epithelia of the female reproductive tract. The mouse fat pad allows transplantation of large tissue fragments, is easily accessible for surgery and imaging, and provides the most favorable native environment for tissues of Müllerian origin.

This protocol describes the primary culture/co-culture of mouse ovarian tissue, using ovaries from neonatal mice and individual ovarian follicles from prepubertal mice. The culture techniques support development in a highly physiological manner, allowing investigation of the effect of extrinsic agents on the ovary, and of interactions between ovarian follicles.
