Spermatid Isolation

Spermatid isolation is the laboratory separation of haploid male germ cells from testicular tissue or mixed germ-cell populations, enabling direct study of late spermatogenesis and male gamete development. It typically combines tissue dissociation with selective separation based on cell size, density, morphology, or molecular markers, while carefully maintaining cell viability and developmental characteristics. Isolated spermatids support research on meiosis, spermiogenesis, chromatin remodeling, gene expression, and cellular differentiation within developmental biology. The approach can also provide defined cell populations for microscopy, molecular analysis, and investigations of how genetic or environmental factors influence the formation of mature spermatozoa.

Step-specific Sorting of Mouse Spermatids by Flow Cytometry

We describe a sorting strategy for mouse spermatids using flow cytometry. Spermatids are sorted into four highly pure populations, including round (spermiogenesis steps 1-9), early elongating (spermiogenesis steps 10-12), late elongating (spermiogenesis steps 13-14) and elongated spermatids (spermiogenesis steps 15-16). DNA staining, size and granulosity are used as selection parameters.

We describe the isolation of cardiac extracellular matrix from C57Bl/6J control mice, tight-skin mice, and tight-skin mice treated with the IRF5 inhibitory peptide. We also describe the vasodilation studies on the isolated vessels from C57Bl/6J, tight-skin mice and tight-skin mice treated with the IRF5 inhibitory peptide.
