Peptide Mass Fingerprinting

Peptide mass fingerprinting is a protein-identification method that uses a characteristic pattern of peptide masses to determine a protein’s identity. In a typical workflow, proteins are enzymatically digested, often with trypsin, the resulting peptides are measured by mass spectrometry, and the mass list is compared with theoretical peptide patterns in sequence databases. In environmental research, this approach supports the identification of proteins from microorganisms and environmental samples, helping characterize microbial communities and biochemical responses. Its results can complement other proteomic and analytical methods, linking molecular composition to ecosystem processes and environmental change.

Species Determination and Quantitation in Mixtures Using MRM Mass Spectrometry of Peptides Applied to Meat Authentication

We present a protocol for identifying and quantifying the components in mixtures of species possessing similar proteins. Mass spectrometry detects peptides for identification, and gives relative quantitation by ratios of peak areas. As a tool food for fraud detection, the method can detect 1% horse in beef.

This protocol shows how to obtain a mass spectrometric "fingerprint" of leukocyte cardiolipin for the diagnosis of Barth syndrome. The assessment of elevated monolysocardiolipin to cardiolipin ratio discriminates patients with Barth syndrome from control heart failure patients with 100% sensitivity and specificity.
